Flavonoids from Rhizophora conjugata fruit extract blocks virulence factors of Pseudomonas aeruginosa

dc.description.abstract Pseudomonas aeruginosa is a major nosocomial pathogen which causes hospital acquired infections and recently has gained importance as a model to study antibiotic resistance. In the present study, we investigated the effect of methanol and methanol: chloroform (1:1) extracts of 7 mangrove plants on P. aeruginosa ATCC 27853 motilities (swarming, swimming and twitching). Amongst the 22 extracts tested, methanolic extract of Rhizophora conjugata fruit showed maximum inhibition. The butanol (RcBu) fraction of this extract almost completely inhibited swarming and swimming motility by 86% each and twitching by 90%, at a concentration of 100mug mL<sup>-1</sup>. This fraction also showed maximum reduction in the production of pyocyanin with 57% inhibition followed by aqueous (RcAq) fraction with 46%. The wetness of the P. aeruginosa colony was reduced in the presence of RcBu and RcAq fractions with percentage inhibition of 64% and 60%, respectively. However, none of the fractions showed significant reduction in rhamnolipid production. The metabolites present in the most active fraction were identified as polymeric flavan-3-ol of catechin/epicatechin and glucoside flavonoid, rutin (2-6) using electrospray ionizationtandem mass spectrometry (ESI-MS/MS). The isolation of (+)- epiafzelechin (1) from of ethyl acetate (RcEA) fraction is also reported here for the first time.
